从微泡菌属AG1(Microbulbifer sp. AG1)克隆得到1302 bp大小的琼胶酶基因,该基因编码产物为一个成熟蛋白(413个氨基酸残基)外加一个信号肽(20个残基)。将不含信号肽片段的琼胶酶在E. coli BL21 (DE3)中进行了异源表达和纯化。使用琼脂糖作为底物,该重组琼胶酶的最适反应温度和pH分别为60℃和7.5。该重组酶表现出优良的热稳定性,在50℃和60℃下处理1 h,重组琼胶酶仍能分别保持67%和19%的残余酶活力。除了SDS,重组琼胶酶对于其他测试的抑制剂、去垢剂和尿素变性剂有着较好的抗性。利用薄层色谱和以对硝基苯-α/β-D-吡喃半乳糖苷为底物的酶活力分析结果表明,该重组琼胶酶为β型琼胶酶,它水解琼脂糖的主要终产物为新琼四糖,而且不同聚合度的酶解产物具有抗氧化活性。  相似文献

稻壳灰与土体混合应用,一方面废弃资源再利用,环保,又可增强土体强度。通过三轴试验,研究不同比例稻壳灰混合黏土及其加筋土应力?应变性能、强度特性以及不同应变水平下模量、偏应力及加筋强度比等土体变化特征。试验结果表明,随稻壳灰比例增加,混合土最大干密度显著减小,最优含水率显著增加。添加不同比例稻壳灰对加筋土抗剪强度有较大影响,10%~15%稻壳灰比例下,加筋稻壳灰混合土初始切线模量和应力峰值达到最大,抗剪强度较优。与土工织物加筋稻壳灰混合土相比,土工格栅加筋稻壳灰混合土偏应力及抗剪强度更大,土工格栅层数对土体抗剪强度增大效果更明显,对应的应力?应变曲线拐点也更突出。试样弹性模量与稻壳灰比例及筋材种类、层数有关,加入稻壳灰后,土体弹性模量增长显著,土工格栅加筋稻壳灰混合土较优比例下可增加1.5倍多,稻壳灰及筋材均能有效提高土体强度。随加筋层数增加,稻壳灰混合土加筋强度比明显增大,与围压关系较小。  相似文献

Based on regional geological setting, stratigraphic distribution and other geological conditions, this paper summarized three types of geothermal reservoirs in the southeast coastal areas of China: Cenozoic sandstone or sandy conglomerate reservoir, Mesozoic granite fissure reservoir and Paleozoic karst reservoir. Cenozoic sandstone or sandy conglomerate reservoirs are mainly located in Cenozoic basins, such as Zhangzhou, Fuzhou, Sanshui and Leiqiong basins. The Tertiary sedimentary basins such as Leiqiong Basin and Sanshui Basin, are controlled by NE-trending faults, while the Quaternary sedimentary such as Zhangzhou and Fuzhou basins are controlled by NW-trending faults. Mesozoic granite fissure reservoirs are mainly distributed in the southeast coastal areas, such as Zhangzhou, Fuzhou, Fengshun, Yangjiang and southern part of Hainan Province. The distribution of good Mesozoic granite fissure reservoir in these areas is mainly controlled by NE-trending faults. Paleozoic carbonate reservoirs are widely distributed in these areas. Most carbonate rocks are from the upper Paleozoic strata, such as those in the area of Huizhou in Guangdong Province. The major types of geothermal systems in the southeast coastal areas of China belong to medium and low-temperature convection. The geothermal resources developed from the ground to-3 000 m underground could be utilized directly for space heating, greenhouse heating, aquaculture pond heating and industrial uses, as well as other purposes. The geothermal resources with a depth of 3 000~6 000 m underground is mainly featured by Hot Dry Rock(HDR) with a temperature ranges from 150 ℃ to 200 ℃, which is conductive to the development of Enhanced Geothermal System(EGS) and can be utilized for power generation.  相似文献

探究微藻的氮代谢通路对了解其对不同氮源利用的分子机制具有重要意义。本研究利用Illumina高通量测序技术对两种氮素营养条件下(硝酸氮和尿素)多形微眼藻的转录组进行分析,通过基因功能注释及数字基因表达谱分析,研究了多形微眼藻细胞内氮代谢的调控机制。结果检测出15种参与氮代谢的酶,对应76个编码基因,构建了多形微眼藻的氮代谢通路图。其中10个酶编码基因在两种不同氮素营养条件下存在差异表达,最显著的是谷氨酸合成酶、谷氨酸脱氢酶和谷氨酰胺合成酶相关基因。有机氮源(尿素)实验组中,多形微眼藻细胞内的硝酸盐还原酶、亚硝酸盐还原酶等基因的差异表达明显高于无机氮源(硝酸钠)实验组,表明当环境中的氮源为尿素时,会对多形微眼藻细胞内硝酸盐的转化和利用有一定影响。本研究初步阐述了硝酸盐、尿素的吸收转运对多形微眼藻细胞内氮代谢的影响机制,可为硅藻在不同氮素营养条件下的吸收利用机制及氮代谢响应研究提供依据。  相似文献

先进天基太阳天文台(Advanced Space-based Solar Observatory, ASO-S)卫星是我国首颗太阳观测卫星, 主要观测太阳耀斑和日冕物质抛射以及产生它们的磁场结构. ASO-S卫星的科学应用系统是科学卫星工程的6大系统之一, 它连接科学用户和卫星数据, 为将卫星的科学数据转化为科学成果提供保障. 科学应用系统的数据库是连接软件与海量数据的枢纽, 为科学数据生产和用户服务及运行提供数据层的支撑. 介绍了科学应用系统的数据库架构设计、数据库的选择以及数据库性能优化和表样例. 这里的数据库包括观测计划、工程参数、运维日志、科学数据、定标数据和特征事件识别等数据库. 这些数据库的建设将为ASO-S卫星工程科学应用系统的顺利运行提供数据支撑, 也可以为未来其他科学卫星类似数据库的搭建提供参考和借鉴.  相似文献

To investigate the sources of particulate organic matter (POM) and the impact of Three Gorges Dam (TGD), two large lakes and erosion processes on determining the composition and flux of POM in low water discharge periods along the middle and lower Changjiang, suspended particulate samples were collected along the middle and lower reaches of the Changjiang (Yangtze River) in January 2008. Organic geochemistry of bulk sediment (particulate organic carbon, organic carbon to nitrogen molar ratio (C/N), stable carbon isotope (δ13C) and grain size) and biomarker of bulk sediment (lignin phenols) were measured to trace the sources of POM. The range of C/N ratios (6.4–8.9), δ13C (?24.3‰ – ?26.2‰) and lignin phenols concentration Λ8 (0.45 mg/100 mg OC‐2.00 mg/100 mg OC) of POM suggested that POM originated from the mixture of soil, plant tissue and autochthonous organic matter (OM) during the dry season. POM from lakes contained a higher portion of terrestrial OM than the mainstream, which was related to sand mining and hydropower erosion processes. A three end‐member model based on δ13C and Λ8 was performed. The results indicated that soil contributed approximately 50% of OM to the POM, which is the dominant OM source in most stations. POM composition was affected by total suspended matter (TSM) and grain size composition, and the direct OM input from two lakes and channel erosion induced OM. The lower TSM concentration in January 2008 was mainly caused by seasonal variations; the impact from the TGD in the dry season was relatively small. A box model indicated that more than 90% of the terrestrial OM transported by the Changjiang in January 2008 was from the middle and lower drainage basins. Northwest Africa (NWA) 4898 is the only low‐Ti, high‐Al basaltic lunar meteorite yet recognized. It predominantly consists of pyroxene (53.8 vol%) and plagioclase (38.6 vol%). Pyroxene has a wide range of compositions (En12–62Fs25–62Wo11–36), which display a continuous trend from Mg‐rich cores toward Ca‐rich mantles and then to Fe‐rich rims. Plagioclase has relatively restricted compositions (An87–96Or0–1Ab4–13), and was transformed to maskelynite. The REE zoning of all silicate minerals was not significantly modified by shock metamorphism and weathering. Relatively large (up to 1 mm) olivine phenocrysts have homogenous inner parts with Fo ~74 and sharply decrease to 64 within the thin out rims (~30 μm in width). Four types of inclusions with a variety of textures and modal mineralogy were identified in olivine phenocrysts. The contrasting morphologies of these inclusions and the chemical zoning of olivine phenocrysts suggest NWA 4898 underwent at least two stages of crystallization. The aluminous chromite in NWA 4898 reveals that its high alumina character was inherited from the parental magma, rather than by fractional crystallization. The mineral chemistry and major element compositions of NWA 4898 are different from those of 12038 and Luna 16 basalts, but resemble those of Apollo 14 high‐Al basalts. However, the trace element compositions demonstrate that NWA 4898 and Apollo 14 high‐Al basalts could not have been derived from the same mantle source. REE compositions of its parental magma indicate that NWA 4898 probably originated from a unique depleted mantle source that has not been sampled yet. Unlike Apollo 14 high‐Al basalts, which assimilated KREEPy materials during their formation, NWA 4898 could have formed by closed‐system fractional crystallization.  相似文献

Bulk major element composition, petrography, mineralogy, and oxygen isotope compositions of twenty Al‐rich chondrules (ARCs) from five CV3 chondrites (Northwest Africa [NWA] 989, NWA 2086, NWA 2140, NWA 2697, NWA 3118) and the Ningqiang carbonaceous chondrite were studied and compared with those of ferromagnesian chondrules and refractory inclusions. Most ARCs are marginally Al‐richer than ferromagnesian chondrules with bulk Al2O3 of 10–15 wt%. ARCs are texturally similar to ferromagnesian chondrules, composed primarily of olivine, pyroxene, plagioclase, spinel, Al‐rich glass, and metallic phases. Minerals in ARCs have intermediate compositions. Low‐Ca pyroxene (Fs0.6–8.8Wo0.7–9.3) has much higher Al2O3 and TiO2 contents (up to 12.5 and 2.3 wt%, respectively) than that in ferromagnesian chondrules. High‐Ca pyroxene (Fs0.3–2.0Wo33–54) contains less Al2O3 and TiO2 than that in Ca,Al‐rich inclusions (CAIs). Plagioclase (An77–99Ab1–23) is much more sodic than that in CAIs. Spinel is enriched in moderately volatile element Cr (up to 6.7 wt%) compared to that in CAIs. Al‐rich enstatite coexists with anorthite and spinel in a glass‐free chondrule, implying that the formation of Al‐enstatite was not due to kinetic reasons but is likely due to the high Al2O3/CaO ratio (7.4) of the bulk chondrule. Three ARCs contain relict CAIs. Oxygen isotope compositions of ARCs are also intermediate between those of ferromagnesian chondrules and CAIs. They vary from ?39.4‰ to 13.9‰ in δ18O and yield a best fit line (slope = 0.88) close to the carbonaceous chondrite anhydrous mineral (CCAM) line. Chondrules with 5–10 wt% bulk Al2O3 have a slightly more narrow range in δ18O (?32.5 to 5.9‰) along the CCAM line. Except for the ARCs with relict phases, however, most ARCs have oxygen isotope compositions (>?20‰ in δ18O) similar to those of typical ferromagnesian chondrules. ARCs are genetically related to both ferromagnesian chondrules and CAIs, but the relationship between ARCs and ferromagnesian chondrules is closer. Most ARCs were formed during flash heating and rapid cooling processes like normal chondrules, only from chemically evolved precursors. ARCs extremely enriched in Al and those with relict phases could have had a hybrid origin (Krot et al. 2002) which incorporated refractory inclusions as part of the precursors in addition to ferromagnesian materials. The occurrence of melilite in ARCs indicates that melilite‐rich CAIs might be present in the precursor materials of ARCs. The absence of melilite in most ARCs is possibly due to high‐temperature interactions between a chondrule melt and the solar nebula.  相似文献

通过对新疆柯坪地区中、上奥陶统萨尔干组和坎岭组高分辨率磁化率数据进行旋回地层学研究,识别出405kyr 长偏心率,111 kyr 短偏心率,38.6kyr、34.1 kyr 和31.0kyr 斜率,以及21.8kyr、20.2 kyr、18.5 kyr 和18.2 kyr岁差旋回,其中斜率信号突出.滤波结果显示萨尔干组和坎岭组记录了 13个405 kyr长偏心率沉积旋回.通过天文校准得出萨尔干组的沉积时限是~2.63 Myr,沉积速率为4.94 m/Myr;坎岭组的沉积时限是~2.64 Myr,沉积速率为5.30m/Myr.萨尔干组滞流盆地相页岩比坎岭组陆棚灰岩沉积速率低.研究层段的0.16-0.17 m沉积旋回信号明显,表明地轴斜率是研究区中、晚奥陶世重要的气候驱动因子.旋回地层学研究结果还赋予生物化石带地质时代意义,大湾沟剖面所产牙形石带Eoplacognathus suecicus、Pygodus serra、Pygodus anserinus、Baltoniodus variabilis 和 Baltoniodus alobatus 的综合延续时限大于5.27 Myr;所产笔石带 Pterograptus elegans、Glossograptus hincksii、Nemagraptus gracilis 和 Corynoides americanus 的综合延续时限为~5.27 Myr.  相似文献
